چکیده

Several studies have been made of the oxidation of C, compounds by micro-organisms grown on such compounds as sole source of carbon. In some cases, rather different results have been obtained by different workers. Kaneda & Roxburgh (1959) obtained evidence for an NAD+-linked methanol dehydrogenase in methanol-grown Pseudomonas PRL-W 4, whereas neither Harrington & Kallio (1960) nor Anthony & Zatman (1963) found such an enzyme in methanol-grown Pseudomonas methanica or Pseudlomonas sp. M 27 respectively. Instead, Harrington & Kallio ( 1960) suggested that methanol was oxidized by P. methanica '...via a catalaselinked peroxidase to formaldehyde which in turn is oxidized by a substrate specific aldehyde dehydrogenase requiringNADI and GSH. . . '. Anthony & Zatman (1963) showed by inhibitor experiments that methanol oxidation by Pseudomonas sp. M 27 was independent of catalase and instead found a methanol dehydrogenase that could be linked to phenazine methosulphate. It is somewhat unexpected that such closely similar bacteria should display different oxidation pathways for methanol, and to furnish further information on this question we present below a study of the oxidation of C1 compounds by methanol-grown Pseudomonas AM 1 and a comparative study of the oxidation of C1 compounds by three other types of bacteria.

Pathways Leading to and from Serine during Growth of Pseudomonas AMI

1. The following enzymes of the phosphorylated pathway of serine biosynthesis have been found in methanoland succinate-grown Pseudomona8 AM 1: phosphoglycerate dehydrogenase, phosphoserine-a-oxoglutarate aminotransferase and phosphoserine phosphohydrolase. Their specific activities were similar in the organism grown on either substrate. 2. A procedure for preparation of auxotrophic mutants of P...

Physiological studies of methane and methanol-oxidizing bacteria: oxidation of C-1 compounds by Methylococcus capsulatus.

Methylococcus capsulatus grows only on methane or methanol as its sole source of carbon and energy. Some amino acids serve as nitrogen sources and are converted to keto acids which accumulate in the culture medium. Cell suspensions oxidize methane, methanol, formaldehyde, and formate to carbon dioxide. Other primary alcohols are oxidized only to the corresponding aldehydes. Oxidation of formate...

Microbial growth on C1 compounds. Incorporation of C1 units into allulose phosphate by extracts of Pseudomonas methanica.

1. Incubation of cell-free extracts of methane- or methanol-grown Pseudomonas methanica with [(14)C]formaldehyde and d-ribose 5-phosphate leads to incorporation of radioactivity into a non-volatile product, which has the chromatographic properties of a phosphorylated compound. 2. Treatment of this reaction product with a phosphatase, followed by chromatography, shows the presence of two compoun...

Aspects of glycine and serine biosynthesis during growth of Pseudomonas AM1 on C compounds.

1. Methanol or formate can replace serine or glycine as supplements for growth on succinate of the auxotrophic mutants 20S and 82G of Pseudomonas AM1, showing that the organism can synthesize glycine and serine in net fashion from C(1) units. 2. Double mutants of Pseudomonas 20S and 82G have been prepared (20ST-1 and 82GT-1) that are unable to grow on succinate+1mm-glyoxylate, succinate+2mm-met...

Microbial Growth on C 1 Compounds 1 . ISOLATION AND CHARACTERIZATION OF PSEUDOMONAS

It has long been known that some non-photosynthetic micro-organisms can grow on organic C, compounds as their sole source of energy and carbon. The means whereby such organisms grow has been the subject of speculation. It has been suggested (Bhat & Barker, 1948; van Niel, 1954) that they might couple the energy of oxidation of the organic substrate to the reduction of carbon dioxide and the syn...
